摘要:本实验采用复合酶法研究了山药多糖的酶配比和提取工艺条件,采用Sevag法脱蛋白分离纯化山药多糖,用薄层层析法研究其组成分。实验结果表明:复合酶用量最佳配比为纤维素酶1.5%、糖化酶1.2%、酸性蛋白酶0.5%,此条件下的山药多糖提取率为2.48%;复合酶法提取最佳工艺条件为提取时间40min、提取温度45℃、料液比1:25,在此条件下,山药多糖提取率为3.12%;山药粗多糖经脱蛋白后,纯度提高了36.93%;通过薄层层析,确定最佳水解酸为硫酸,最佳展开系统为正丁醇:乙酸:水= 4 :1: 5,山药多糖由葡萄糖、甘露糖、果糖组成。

Abstract: In our experiment, we took composite enzymatic to study yam polysaccharide enzyme ratio and extraction conditions, using the method of Sevag to deproteinize separation and purification yam polysaccharide, and the thin layer chromatography study its constituent. The experimental results showed that: the best ratios for composite enzyme dosage were that cellulose enzyme was 1.5%, that saccharifying enzyme was 1.2%, and that acid protease was 0.5%, the conditions of the yam polysaccharide extraction yield was 2.48%.The optimal conditions for composite enzymatic extraction were that extraction time was 40min, and that extraction temperature was 45℃, that feed liquid ratio was 1:25. Under these conditions, the extraction rate of the yam polysaccharide highest total sugar yield of 3.12%.Yam crude polysaccharide increased the purity of the 36.93% through removing protein. By thin layer chromatography, to determine the optimum hydrolysis of the acid was sulfuric acid, the best expand system was n-butanol: acetic acid: water = 4: 1: 5, yam polysaccharide consisted of glucose, mannose and fructose.
