摘  要:[目的] 本研究旨在对放线菌S10A10降解纤维素的筛选及其产酶条件的研究,充分利用纤维素酶降解纤维素的优点,对纤维素的再生利用进行的一系列研究。 [方法] 本文利用CMC透明圈分离法从蝎子体内分离出10株能在以纤维素(CMC-Na)为唯一碳源的筛选培养基上生长的菌株,采用刚果红溶液染色法作进一步识别,获得透明圈最大的一株菌株,命名为S10A10。在对S10A10发酵条件优化中,采用单因子实验法,探讨不同种龄的菌液对发酵的影响,以及酶作用的最适反应温度和最适反应pH。[结果] 采用刚果红染色法,能获得透明圈较大的菌株;在对S10A10产酶条件优化中,该放线菌在发酵12天的酶活力最高,酶作用的最适温度为70 ℃,酶作用的最适pH为3.2。 [结论] 该放线菌是一株嗜酸嗜热的放线菌,产酶活力较高。

Abstract:[Objective] The aims of this study on Actinomyces S10A10 degradation of cellulose by screening and enzyme producing conditions of study, make full use of the advantages of cellulose degradation by cellulase, cellulose regeneration using a series of research. [Method] In this study, ten strains were obtained by a rapid choosing method from scorpions, which could use CMC-Na as the exclusive carbon source. One strain was gained by Congo red dyeing and named S10A10. Optimized zymolytic conditions of S10A10 by single-factorial experiments, such as inoculum age and studied the optimal temperature and PH for reaction of cellulose. [Result] Using Congo red staining method, can obtain the transparent circle larger strains; the S10A10 fermentation conditions optimization, the actinomycete fermentation in the 12 days of the highest enzyme activity, enzyme the optimum temperature of 70 degrees C, enzyme activity for optimum pH 3.2. [Conclusion] The actinomycosis is an acidophilic thermophilic actinomycetes, higher lipase activity.
