摘要:本实验通过单因素试验和正交试验对醋化醋杆菌产乙醛脱氢酶发酵条件进行了优化,得到的醋化醋酸杆菌产乙醛脱氢酶的最适碳源为麦芽糖,最适氮源为酵母浸膏,最适无机盐为磷酸二氢钾,最适碳源浓度为3%,最适氮源浓度为2%,最适乙醇浓度为4%,最适磷酸二氢钾浓度为0.05 g/L。用最优组合进行试验,发现第四天酶活最高,且最高酶活为0.788 U/mL。

Abstract:Fermentation conditions of Acetobacter aceti to produces acetaldehyde dehydrogenase were optimized by single factor experiments and orthogonal experiments, the optimal conditions to produce acetaldehyde dehydrogenase were gotten as follows: the optimal carbon source was maltose, the best nitrogen source for yeast extract, the optimal inorganic salt was potassium dihydrogen phosphate, the optimum carbon source concentration was 3%, the optimum nitrogen source concentration was 2%, the optimal concentration of ethanol was 4%, the optimal potassium dihydrogen phosphate concentration was 0.05 g/L. Under the optimal conditions, the highest enzyme activity was 0.788 U/mL after 4d’ fermentation.
